题目1:Motor neuron disease: Role of posttranslational modifications of SOD1
内容简介:Motor neuron disease, Pathologically it is characterized by loss of motor neurons in the cortex, brainstem, and spinal cord, leading to progressive muscle weakness, atrophy, and death. Over the past two decades a major breakthrough in ALS research is the discovery that mutations in the gene SOD1 are a cause of familial ALS. Strong evidence supports that the familial ALS-linked SOD1 variants are readily susceptible to posttranslational modifications, and subsequently become misfolded. Therefore, selective knockdown of misfolded SOD1 could be a cure for ALS. Building on a peptide-directed lysosomal protein degradation strategy, we have recently succeeded in selectively knocking down of misfolded SOD1. We are hopeful that this strategy may eventually lead to a cure for ALS.
